That should work just fine - at least as long as you either:
1. You use plain DOS with no "emm386" or similar "big memory" features. 
2. Have an AMD processor that supports real-mode in virtual machine -
that way, emm386 and such should work too (emm386 uses protected mode to
map memory at high addresses and then reverts back to real-mode, and
that's tricky business on Intel processors).
