pygrub isn't intended to work on the command line.
I don't agree, pygrub is very usefull to test a device (virtual or not) for grub installation.
use it as the bootloader option in your domU configuration file.
i.e.
bootloader='/usr/bin/pygrub'
FYI: in Xen 3.3 pygrub is replaced with PV-GRUB to improve security.
